Phage Lambda fragment 5 - recombination
This bio-brick is one of eight correlated bio-bricks – each one contains a one fragment of the phage lambda CI875s7. The phages fragments created in order to allow manipulation in the phage genome by manipulation smaller fragments, and re-factor the complete 48kb genome after the manipulation.
Each fragment was inserted into pSB1C3_MCS plasmid (BBa_K784023), which was designed mainly for this purpose. The MCS contain PacI and SpeI recognition sites, which does not appear at the phage genome.

Fragment 5 contains the recombination area that allows the phage lysogenic life cycle (penetration to the host genome). This part includes the att-λ sequence which allows site specific recombination into an E. coli genome (with the matching att-B sequence) between gal and bio operons.
